HOW THYROID HORMONES ENTER TARGET TISSUES: THYROID HORMONE TRANSPORTERS AS CRITICAL GATE-KEEPERS
Um ihre Wirkung zu entfalten, müssen Schilddrüsenhormone in ihre Zielzellen aufgenommen werden. Diese Aufnahme wird von sog. Schilddrüsenhormon (SDH)-Transportern vermittelt. Eine beeinträchtigte Funktion kritischer SDH-Transporter führt zu gewebespezifischen Veränderungen im SDH-Status mit weitreichenden Konsequenzen. So zeigen Patienten mit inaktivem SDH-Transporter MCT8 eine schwere Form der psychomotorischen Retardierung, die durch eine Unterversorgung des Gehirns mit SDH bedingt ist, während gleichzeitig anomale SDH-Konzentrationen in der Zirkulation eine hyperthyreote Situation in der Leber bedingen (sog. Allan-Herndon-Dudley Syndrom). Mit Hilfe von Mausmodellen untersuchen wir die (patho-) physiologische Funktion von MCT8 und anderen SDH-Transportern im Gehirn und peripheren Organen. Darüber hinaus ist es unser Ziel, neue Therapie-Ansätze für Patienten mit SDH-Transporter-Mutationen zu entwickeln.
Thyroid hormone (TH) transporters are mandatory for proper TH action as they mediate the transmembrane passage of TH and can therefore act as gate-keepers in all target cells. A compromised TH transport can lead to pronounced changes in the cell- and tissue-specific TH status that in turn affect the whole organism. For instance, patients with inactivating mutations in the TH transporter MCT8 show a severe form of psychomotor retardation due to low TH concentrations in the brain while abnormal serum TH parameters cause a thyrotoxic situation in liver and other organs (Allan-Herndon-Dudley syndrome). By taking advantage of mouse mutants we aim to define the (patho-) physiological role of MCT8 and other TH-transporters in the CNS as well as peripheral organs and to develop novel therapeutic strategies for patients with TH transporter mutations.
